The project aims to characterize at the molecular level, using single-nucleus multiomics and spatial-omics, the molecular signature that causes muscle loss during tumor growth, also known as cancer cachexia. The molecular profile will be studied in both mouse models and biopsies from cachexia patients. The ultimate goal is to develop an RNA therapy that can counteract tumor-induced gene expression changes and prevent cachexia. The RNA therapy will be tested both in mouse models of cachexia and in patient-derived neuromuscular organoids. It involves the set up of the spatial proteomics on muscle tissue sections from patient biopsies and from muscles of cachectic mice. Spatial proteomics data will then be integrated with spatial transcriptomics data to confirm gene expression changes at the protein level.
