The CLIN-TCR project aims to demonstrate the clinical relevance of the MASCARA platform, a technology developed to scalably and unbiasedly pair T cell receptors (TCRs) with their cognate HLA-presented antigens. The research appointment will contribute to validating the platform in two translational settings: oncology, through the identification of TCR–neoantigen pairs in colorectal cancer samples, and autoimmunity, through the mapping of autoreactive or cross-reactive TCRs in psoriasis. Activities will include the generation of synthetic cell libraries, enzymatic DNA barcoding reactions, sorting, single-cell sequencing, bioinformatic analysis and functional validation of the identified ligands. The programme will generate clinically relevant datasets, draft SOPs and information supporting intellectual property consolidation and future exploitation of the technology.
