Description
The morphological classification of the leafhopper vectors of Grapevine flavescence dorée phytoplasma is often complex and requires extensive entomological expertise. For this reason, the aim of this project is to apply identification protocols based on molecular methods, which represent a more reliable alternative tools for a correct species identification. The use of molecular markers, such as mitochondrial genes, ribosomal genes, or microsatellites, will allow for accurate discrimination of vector species even in the absence of diagnostic morphological characteristics. Sequencing of a portion of the mitochondrial region of the cytochrome oxidase I (COI) gene will be used to confirm suspected morphotypes and to genetically trace the vectors.Furthermore, the mechanisms regulating the interaction between the phytoplasma, the host plant, and the insect vector will be investigated using quantitative PCR (qPCR), gene sequencing, and transcriptomic analysis.
