Description

Title of the project:“Bioenergetic studies in vivo and in vitro models of Troyer Syndrome”.Troyer syndrome (TS) is an autosomal recessive form of Hereditary Spastic Paraplegia (HSP) caused by loss-of-function mutations in SPART gene, which encodes for Spartin, a multifunctional protein. Despite its presence in almost all cell compartments, the key role of Spartin remains elusive, although a role in lipid droplets turnover and mitochondrial homeostasis has been highlighted. We have already demonstrated an alteration in the mitochondrial-associated membrane (MAM) complex and impaired mitochondrial import of nuclear-encoded proteins in two cell models of TS, including the proteins involved in the Coenzyme Q10 (CoQ) biosynthesis. The project aims to investigate the rescue effect of restoring MAMS stability by sphingomyelinase (SMase)..Omissis.
