In Vitro Assessment of Scaffold Cytotoxicity and Biological Efficacy.

The project aims to evaluate the in vitro cytotoxicity and bioactivity of novel biomimetic scaffolds. Cell culture models will be used to perform both direct and indirect cytotoxicity assays assessing cell viability, cell death, and material-induced morphological alterations. The bioactivity phase will focus on the ability of the scaffolds to promote osteochondral cell adhesion, proliferation, and differentiation through the evaluation of extracellular matrix deposition and tissue-specific marker expression using morphological, immunohistochemical, and molecular biology techniques. During the 12-month project, experimental protocols will be optimized, data will be collected and statistically analyzed, and technical-scientific reports will be prepared. The study will be conducted in accordance with the principles of the 3Rs (Replacement, Reduction, and Refinement), contributing to the preclinical validation of novel scaffolds for regenerative medicine applications.
